Abstract

In the pharmaceutical industry there is a constant demand for new drugs. A major challenge is the bioavailability enhancement of the drug molecules. Here the production of Solid Dispersions on a co-rotating twin screw extruder via Hot-Melt-Extrusion is a promising method. [1]

Description

Twin-Screw-Extrusion offers the advantage that the process combines several unit operations in one apparatus, e.g. mixing, melting and dissolving. The screw has a modular design with for example mixing, conveying and kneading elements. Through the combination of different unit operations the process is difficult to optimize and a challenge regarding the scale up. Computer based simulations are a good possibility to predict critical process parameters like residence time, pressure and temperature. [1] Based on the calculations from Kohlgrüber a 1D model was developed. Important inputs for this model are dimensionless parameters that characterize pressure and power correlations for every screw element. [2]

The aim of this study is to determine these parameters and thus to design the extrusion process through specifically selected screw configurations. For the determination a precise knowledge of the basic operations in an extruder is indispensable.
