Abstract

One of the major problems for the development of new drug substances is the poor aqueous solubility. New and innovative formulation techniques to increase the dissolution rate as well as the solubility of the drug need to be researched in order to enhance their respective bioavailability.

Description

Solid crystalline suspensions (SCS) are a promising formulation for BCS II drug substances. This type of solid dispersion consists of a water-soluble continuous phase, also called matrix, and dispersed drug particles. Since both components are in crystalline form, the final product is thermodynamically stable offering a major advantage over amorphous solid dispersions regarding shelf life.

In this work a process for the production of SCS in a technical scale is developed. Major goal is to achieve small drug particle sizes in the target range of 0.1-1µm and dispersing them inside the matrix in a single-step process.

This is a research project in cooperation with INVITE GmbH.
